Safety summary: Ipamorelin

In short: In the ileus trial adverse events were common in both groups (87.5% ipamorelin vs 94.8% placebo), largely reflecting surgery. Long-term safety of repeated use for muscle or "anti-aging" has not been studied in controlled trials.

Regulatory Warning

Not an FDA-approved drug for any medical condition. Prohibited in sport under WADA Category S2.

Most common side effects

Reported symptoms in clinical trials and published pharmacology literature:

Symptom Reported rate Clinical context & notes
Mild headache Common in clinical studies Transient, usually resolving within hours.
Flushing & lightheadedness Common Mild vasodilation occurring shortly after injection.
Increased appetite Mild (much less than GHRP-6) Acts on the ghrelin receptor, but with greater selectivity than earlier secretagogues.
Injection site discomfort Common Mild localized stinging or redness.

Serious risks and warnings

Potential adverse reactions and biological risks identified in research:

  • Impaired glucose tolerance: Elevated growth hormone levels can reduce peripheral insulin sensitivity over prolonged periods.
  • Water retention & joint discomfort: Transient soft-tissue swelling from elevated GH and IGF-1.
  • Unregulated product contamination: Compounded or online research vials carry risks of microbial and endotoxin exposure.

Contraindications

This compound should not be used in individuals with:

  • Active cancer
  • Pregnancy or nursing
  • Athletes subject to anti-doping testing

Common questions about Ipamorelin safety

Does ipamorelin raise cortisol or prolactin?

Unlike GHRP-2 and GHRP-6, clinical pharmacology studies in healthy volunteers (PMID 10496658) showed that ipamorelin is selective for growth hormone and does not significantly elevate ACTH, cortisol, or prolactin at standard doses.

Is ipamorelin safe for long-term anti-aging use?

There are no long-term clinical trials evaluating ipamorelin for anti-aging, fat loss, or muscle gain. Its only human trials were short hospital-based studies for bowel motility after surgery.

Can ipamorelin be detected on sports drug tests?

Yes. WADA specifically lists ipamorelin as a prohibited growth hormone secretagogue (Category S2). Anti-doping laboratories routinely detect ipamorelin and its metabolites using LC-MS/MS.
